GLEMITOL PM 2 combines Glimepiride, Pioglitazone, and Metformin in a triple-action formulation designed to manage blood sugar levels in adults with type 2 diabetes mellitus. Glimepiride is a sulfonylurea that stimulates insulin secretion from the pancreas, Pioglitazone is a thiazolidinedione that improves insulin sensitivity, and Metformin decreases hepatic glucose production. This combination targets different aspects of blood glucose regulation, providing comprehensive glycemic control and reducing the risk of diabetes-related complications. GLEMITOL PM 2 is available in 10x10 blister packaging.
HOW DOES IT WORK?
Glimepiride increases insulin release by stimulating pancreatic beta cells, helping lower blood glucose levels, particularly after meals.
Pioglitazone improves insulin sensitivity in the body’s muscle and fat tissues, making it easier for cells to take up glucose and lower blood sugar.
Metformin reduces glucose production in the liver and improves insulin sensitivity, particularly effective in lowering fasting blood glucose levels.
Together, these agents work synergistically to provide both basal and postprandial (after-meal) glucose control.
SIDE EFFECTS
While GLEMITOL PM 2 is generally effective, some patients may experience side effects, such as:
Hypoglycemia (low blood sugar)
Gastrointestinal discomfort (nausea, diarrhea)
Weight gain
Edema (fluid retention)
Headache
Patients are advised to monitor for symptoms of hypoglycemia and consult their healthcare provider if side effects persist or worsen. Pioglitazone may increase the risk of fluid retention and should be used with caution in patients with heart conditions.
ADMINISTRATION OF DOSES
GLEMITOL PM 2 is usually prescribed as one tablet taken once or twice daily with meals to reduce the risk of gastrointestinal discomfort. Dosage adjustments may be required based on individual glycemic control and tolerance. Patients should adhere to their healthcare provider's dosing instructions and avoid skipping or doubling doses.
BENEFITS
Provides effective control over blood sugar levels by addressing multiple mechanisms of glucose regulation(Goodman & Gilman's: The Pharmacological Basis of Therapeutics, 13th Edition)
Reduces the risk of long-term complications of type 2 diabetes with comprehensive glycemic control(Katzung & Trevor's Basic and Clinical Pharmacology, 14th Edition)
INDICATIONS
GLEMITOL PM 2 is indicated for:
Management of type 2 diabetes mellitus in adults when diet and exercise alone are insufficient(Harrison's Principles of Internal Medicine, 20th Edition)
PRECAUTIONS
Use GLEMITOL PM 2 cautiously in patients with kidney or liver impairment, heart failure, or a history of hypoglycemia. Avoid excessive alcohol consumption, as it may exacerbate Metformin’s gastrointestinal effects or lead to hypoglycemia. Pioglitazone may cause or worsen fluid retention, so it is not recommended for patients with severe heart failure. Pregnant or breastfeeding women should consult a healthcare provider before using this medication. Regular monitoring of blood glucose, kidney function, and liver enzymes is recommended.PLEASE NOTEAlways consult your doctor before taking GLEMITOL PM 2 or any diabetes medication.
